Telomerase is a key participant of the telomere length maintenance machinery; it extends 3’-ends of linear chromosomes in eukaryotic cells. This enzyme is a ribonucleoprotein that contains telomerase reverse transcriptase and telomerase RNA component. The processing mechanism of human telomerase RNA involves exosome trimming or degradation of the primary precursor. Recent data provide evidence that the competition between the processing and decay of telomerase RNA may regulate the level of RNA at the physiological level. However, the events that result in the appearance of the primary transcript need to be clarified. To investigate the mechanism of human telomerase RNA processing we developed a reporter system that allowed us to prove the promoter- dependent manner of regulation of the hTR transcription termination. We proposed that the termination of transcription of hTR may involve Integrator, which is a multisubunit complex supposed to participate in the regulation of transcription of noncoding RNAs. With a help of series of different techniques, we have shown that the Integrator complex is indeed a key component in the regulation of promoter-dependent transcription termination of hTR and regulation of further fate of the primary hTR transcript.
