Lysozyme was chosen as a model object for studying its self-organization at different surfaces including aqueous-air, aqueous-nanodiamond, and liquid-liquid interface. In present work we used tritium label as tool for determine both the value of protein concertation at the interface and its structure in the adsorption layer. To determined the amount of lysozyme at the aqueous-organic liquid and aqueous-nanodiamond interfaces tritium labeled protein was used. [3H]Lysozyme adsorption at the liquid-liquid interface was studied by means of scintillation phase method. Lysozyme sample was introduced in the system of two immiscible liquids one of each is a scintillator. The adsorption value was calculated from measured radioactivity. Adsorption of lysozyme on nanodiamond surface from the aquatic solution was determined by radioactivity of the adsorbed protein. The structure of the adsorption layer at both aqueous-air surfaces and protein modified nanodiamond was studied by treatment with the atomic tritium following by total hydrolysis of the protein and amino acids analysis with mass and radioactivity control. Peculiarities of the protein and protein-surfactant mixture self-organization at the interfaces that were revealed by tritium probe approach will be discussed in the presentation.
