Telomerase is a ribonucleoprotein that contains telomerase reverse transcriptase and telomerase RNA component. Reverse transcriptase synthesizes telomeres using the telomerase RNA template region. Telomerase is only active in the subset of human cells, such as stem cells and germ cells. Human telomerase RNA (hTR) is transcribed from its own promoter by polymerase II. The hTR processing mechanism involves exosome trimming or degradation of the primary precursor. Multisubunit complex Integrator is required for 3’-end processing of pre-snRNAs in humans. Integrator is recruited to snRNA promoters and associates with the C-terminal domain of RNA polymerase II. We showed that complex Integrator is involved into termination of transcription of human telomerase RNA followed by further processing. In our work we made a knockdown of putative Integrator scaffold subunit and catalytic subunits in HEK293 cells. The level of total hTR and 3’-end extended transcripts of hTR was analysed by qRT-PCR. We observed an enrichment of the 3’-end extended form of hTR by 2-times in cells treated with siRNAs. Our results mean that Integrator participates in pre-hTR formation.
