Lynx1 is one of endogenous peptides in the brain, belonging to Ly6/uPAR family [1]. Lynx1 has been shown to bind the different types of nicotinic acetylcholine receptors (nACh-receptors) [2]. But it is remained unclear how Lynx1 affects nACh-receptors function in the brain. Here we study the effect of water-soluble recombinant analogue of human Lynx1 (ws-Lynx1) on interneurons of the layer 1 responding to 1mM acetylcholine in the rat visual cortex using whole-cell patch-clamp configuration and fast drug-application system. It was revealed that ws-Lynx1 at 1μM concentration didn’t affect the peak-amplitude ACh-evoked current, while application of 10μM ws-Lynx1 resulted in the ~ 30% enhancement of the peak-current amplitude. Using specific inhibitors 4μM MLA and 8mM DhbE, α7ACh-receptors was identified to be involved in the responses observed. Thus ws-Lynx1 at 10μM, but not at 1μM affects the α7ACh-receptors in L1 interneurons in the rat visual cortex increasing the peak-current amplitude of responses.
