Dermal papilla (DP) cells of hair follicle represent a very useful biological model and may be utilized in practical medicine. We and other scientists have shown that these cells are able to differentiate along osteogenic, adipogenic and neural lineages. They reorganize collagen matrix and induce epidermal morphogenesis and angiogenesis. Unfortunately, DP cells quickly loose their specific hair-inductive ability during cultivation. It correlates with diminished staining for alkaline phosphatase (AP). Addition of certain growth factors, substances, and matrix components prolongs time of AP-positive cultivation. Nevertheless, there is still no ideal culture system for preservation of unique DP properties. It is also unclear how culture conditions lead to the loss of specific DP properties. We compared gene expression in newly isolated and cultivated DP cells to look for changes. We found three to six fold increase in expression of BMP6, FGFR1, and NDP; 1.5-2 fold increase in expression of FGFR2, ALX4, NOG, SOX, HHIP, HEY1, WIF1, LTBP1, and PRSS12. We further performed deep cDNA sequencing using GAIIx Illumina platform. More than 70 genes were found to have expression significantly (3 or more fold) increased in DP cells after passaging and 40 genes to have expression significantly decreased. We divided affected genes into groups regarding their function. Genes involved in cell adhesion formed one of the largest group. Thus, spontaneous epigenetic changes occur in cultivated DP cells. This provides a useful model to compare changes in functional activity and transcriptome of DP cells. The data obtained may help search for optimal culture condition for DP cells.
