Formate dehydrogenase (FDH) occurs in a broad spectrum of living cells: bacteria, fungi and plants. The role of FDH varies in different types of organisms. For pathogenic bacteria and fungi, and plants FDH is considered to be the enzyme of stress response. The amount of the enzyme strongly increases in case of different stress conditions. FDHs from different sources are also of high practical interest, as they can be used for NAD(P)H regeneration in different biotechnological processes. In pathogenic bacterium Staphylococcus aureus FDH is overexpressed when cell are in biofilms. Two possible variants of gene encoding FDH from S.aureus (SauFDH1 and SauFDH2 – long and short forms, respectively) were cloned and expressed in E.coli cells as active and soluble enzymes. SauFDH1 and SauFDH2 were purified and characterized. Long form SauFDH1 showed better kinetic properties and higher thermal stability compared to SauFDH2. Moreover, stability of SauFDH1 was only a little bit lower than one for the recombinant wild-type FDH from Pseudomonas sp. 101 which is one of the most stable formate dehydrogenases. SauFDH1 also showed high chemical stability.
