Разработка методик определения стероидных гормонов в биологических жидкостях человека с использованием хромато-масс-спектрометрических методов анализа и современных способов пробоподготовки
Steroid hormones act as regulators of numerous important processes in humans. In case of their contents change, a risk of development of various diseases exists, therefore there is an urgent need of their control in human biological fluids. Depending on the task, several biological fluids can be used, e.g., saliva and blood allow to obtain real-time concentrations, while urine shows average steroid contents. In terms of sample obtaining, saliva and urine are more preferable, sine the procedure of their obtaining is painless and non-invasive. Nowadays, several methods for their quantification in biological fluids exist. Among them, the most promising technique is chromatography coupled to mass spectrometry. However, owing to trace and ultra-trace steroid contents in biological fluids, sample pretreatment is required to obtain accurate results. For this purpose, traditional sample preparation techniques are used: liquid-liquid and solid-phase extraction. These techniques suffer from many disadvantages including consumption of large amounts of toxic solvents, laboriousness, relatively high analysis cost and, in case of solid-phase extraction, the use of disposable cartridges. Considering the above mentioned shortcomings, the development of fast and accurate procedures for the quantification of steroid hormones in biological fluids is required. Since the studied matrices are complicated, achievement of adequate results becomes possible when using microextraction techniques which do not have the mentioned shortcomings and meet the requirements of green chemistry. As a results of the conducted studies, a set of selective and accurate procedures for the quantification of steroid hormones will be developed allowing to significantly enhance the efficiency of diagnostic as well as doping control laboratories. In the future, a set of solutions related to the increase in the number of simultaneously detected substances will be proposed.
