Аннотация:Opening of Cl- channels in arterial smooth muscle (ASM) leads to Cl- efflux from the cell thereby causing depolarization and contraction. The functioning of ASM changes significantly during maturation. However, whether the regulatory role of Cl- to the contractile responses changes during early postnatal development is not clear, despite maturational changes in Cl- homeostasis were shown previously in neurons. Thus, we hypothesized the role of Cl- in the regulation of arterial contractile responses to differ in young and adult rats. We used the muscular-type saphenous artery from male Wistar rats aged 10-12 days (young) and 3-4 months (adult). All procedures were approved by Russian institutional committee on animal welfare. Endothelium-denuded arterial segments were mounted in isometric myograph (DMT A/S). Contractile responses were evoked by α1-adrenoreceptor agonist methoxamine (10-8-10-4 M). The effects of Cl- removal from the solution (equimolar substitution of NaCl and KCl by respective aspartate salts), NKCC1 blockade (bumetanide, 30 μM), Cl- channels blockade (MONNA, 3 μM) were studied. Removal of Cl- decreased contractile responses to methoxamine, the effects were more prominent in arteries of young rats compared to adults. Bumetanide in normal solution as well as bicarbonate-free solution did not change the contractile responses in either young or adults. MONNA stronger reduced contractile responses in arteries of young animals in comparison to adults. The obtained results demonstrate that Cl- is more important for the arterial contractile responses in 10-12-day old rats than in adult rats. Study was supported by RFBR (grant N 18-015-00216-а).
