Enzymatic methods that were proposed for the determination of alcohols within the past 20 years and based on the use of two enzymes, namely, alcohol dehydrogenase and alcohol oxidase are considered. Alcohol dehydrogenase catalyzes oxidation reactions of ethanol and other aliphatic alcohols (C2-C20) that involve the coenzyme nicotinamide adenine dinucleotide (NAD+) and yield the corresponding aldehyde and the reduced form of nicotinamide adenine dinucleotide (NADH). Alcohol oxidase catalyzes the oxidation of all lower aliphatic alcohols (C1-C6) by air oxygen. Different methods for monitoring the rate of indicator reactions are considered. The rate of reactions catalyzed by alcohol dehydrogenase is monitored by the amount of resulting NADH using spectrometric (spectrophotometric or luminescence) or electrochemical (potentiometric with the use of electrodes at which the enzyme and coenzyme are simultaneously immobilized) methods. The rates of reactions catalyzed by alcohol oxidase are determined either from the amount of oxygen consumed for the oxidation of alcohols (by electrochemical methods using enzyme electrodes based on the Clark electrode) or from the amount of the resulting hydrogen peroxide (by electrochemical methods using enzyme electrodes or by spectrometric methods using sets of consecutive reactions yielding a product that absorbs or fluoresces in the ultraviolet or visual spectral region). Some attention is given to enzymatic test methods for the determination of alcohols.
