Аннотация:The effect of chitosan on epidermal and guard cells in the epidermis isolated from pea leaves was investigated. Chitosan induced the condensation and marginalization of chromatin and the subsequent destruction of nuclei in epidermal, but not in guard cells. The addition of Н2О2 produced a similar effect on the cells. Nitroblue tetrazolium, pyruvate and mannitol, possessing antioxidant properties, inhibited the destruction of nuclei caused by chitosan or Н2О2. The chitosan-induced destruction of epidermal cell nuclei was prevented under anaerobic conditions. Diphenylene iodonium and quinacrine, inhibitors of the NADPH oxidase of the plasma membrane, suppressed the destruction of nuclei induced by chitosan. Chitosan caused the generation of reactive oxygen species and, similar to Н2О2, impaired the permeability barrier of the plasma membrane in guard cells. The data obtained demonstrate that chitosan and Н2О2 produce similar effects on plant cells. Chitosan initiates the production of reactive oxygen species which induce programmed cell death, presumably by activating the NADPH oxidase of the plasma
membrane.
