Аннотация:Introduction: Parkinsonism is characterized by progressive loss and damage of nigral dopaminergic neurons resulting in the reduction of dopamine level in the striatum. Phytomix-40 (Phm-40) is consisting of the forty plant water-ethanol extracts (Panax ginseng, Rhodiola r. L.; Eleutherococcus s.; etc.) and contains ginsenosides (Rbl, Rb2, Rgl, Rg2). eleutherosides, flavonoids, etc. It has been demonstrated that ginsenosides possess neurotrophic and neuroprotective properties. The aim of this work was to study the effect of Phm-40 on parkinsonian syndrome manifestation and caspase-3 activity in substantia nigra of C57BL/6 mice with l-methyl-4-phenyl-l,2,3,6-tetrahydropyridine(MPTP)-induced parkinsonian syndrome (PS).Methods: PhM-40 was administered to mice per os with drinking water in 10%-concentration 10 days following the beginning of MPTP injection. Caspase-3 activity was assayed in substantia nigra extracts by extrication rate of Ac-DEVD-p-nitroanilide (caspase-3 substratum).Results: The administration of PhM-40 resulted in a decrease of parkinsonian symptoms formation (oligokinesia and rigidity) on day 5 and day 10 after the beginning of MPTP injection. MPTP administration caused essential reduction of DA, HVA and DOPAC levels in the striatum and increase of caspase-3 activity in substantia nigra of mice. Chronic administration of PhM-40 in parkinsonian mice reduced caspase-3 activity in substantia nigra and increased the striatal levels of DA, DOPAC and HVA by 46%, in 4.1 and 2.3 times, correspondingly.Conclusion: PhM-40 reduced parkinsonian symptoms formation and partially blocked apoptosis in substantia nigra. It is suggested that PhM-40 possess neuroprotective properties which explains its antiparkinsonian effect in mice.
