Аннотация:A rapid and sensitive immunochromatographic assay (ICA) was proposed to detect herbicide 2,4-dichlorophenoxyacetic acid (2,4-D) in fresh juices. ICA was carried out in an indirect competitive format with sequential incubations of a test strip with a mixture of specific antibodies and the analyte and then, with anti-species antibodies conjugated with gold nanoparticles (GNPs). The assay protocol including concentrations of the 2,4-D-protein conjugate in the test zone, anti-2,4-D antibodies, anti-species antibody-GNPs conjugate, and duration of the assay steps was optimized to achieve a minimal limit of detection (LOD). The optimized ICA was characterized by instrumental/visual LODs of 2,4-D equal to 0.1/10 ng/mL and a working range of detectable concentrations from 0.2 to 5.1 ng/mL. The ICA has been successfully applied to detect 2,4-D in different fresh juices, which are popular drinks among children and adults. A quick sample preparation by simply diluting of the juice with a buffer was proposed. The recoveries of 2,4-D in juices were 84–112 %, which demonstrated reliable determination of the herbicide in real food matrices. The entire testing cycle (from receiving a sample to evaluating the analysis results) took only 25 min.
