Аннотация:The aim of this study was to identify potential markers and mechanism of spontaneous transformation of multipotent stromal cells (MSC) with perivascular immunophenotype.
A transcriptome comparative analysis of 6 paired samples of normal and spontaneously transformed MSCs with perivascular immunophenotype was performed after isolation from adipose tissue of 6 healthy donors at the first passages. According to data obtained by microarray Illumina HT-12 v4, differentially expressed transcripts were identified by Significance Analysis of Microarrays (SAM) and put through statistical analysis using Gene Ontology (GO) database and the Molecular Signatures Database (MSigDB).
The results suggest connection of the spontaneous transformation of MSC with known pathways of oncogenic transformation (E2F, ATR/ATM, RAS, RHOA) and give direction for further investigations. Also a panel of potential markers has been presented, several of which have poorly investigated functions and hasn’t been previously associated with oncology (HSPB6, PLAC9, FEZ1, DTWD1, APH1A, ATP5L).
