Аннотация:The constitutive heterochromatin is defined as a highly compacted transcriptionally inert chromatin. In the current study we focused on transcription and interphase 3-D organization of the human pericentromeric satellite 3 DNA (HS-3) from the locus 1q12 in embryonic and extraembryonic tissues. At 8-11 wks, the locus 1q12 in chorion and embryonic cells is localized within chromocenters close to the nuclear periphery. At the earlier stages (4-5 wks of gestation), the locus HS-3 relocates toward the centrum of the nucleus. HS3 polyadenilated transcipts were found in the embryonic organs and chorion on the 7-9 weeks of gestation. The choice of the chain for transcription depends on the cell type and gestation age. No transcripts were found in chorion and embryos after the week 9. The results confirm HS3 transcription at the early stages of human organogenesis which is accompanied by spatial 1q12 locus rearrangements.
