Аннотация:By modifying the amino group of luminol, 5-(4-hydroxy-1,3-dioxoisoindolin-2-yl)-2,3-dihydrophthalazine-1,4-dione (HDDD) was synthesized. Using a combination of full factorial design and “one-variable-time” method, favorable conditions for the generation of HDDD-dependent chemiluminescence were determined. It was shown that replacing luminol with HDDD in the HRP-C-catalyzed enhanced chemiluminescence reaction increased the luminescence intensity. With HDDD as a substrate of HRP-C the chemiluminescent assays for determination of HRP-C and hydrogen peroxide with improving sensitivity were developed. HDDD was also successfully used in chemiluminescent bi-enzyme system for choline determination using simultaneously HRP-C and choline oxidase. The feasibility of the bi-enzyme assay was proved in the assessment of choline content in milk samples. The obtained results demonstrated good prospects for the wide application of HDDD in the development of new highly sensitive chemiluminescent assays.
